The packs work, the number on hand goes up every time you use one, but you can only use one if a dweller actually has taken damage. Same for the radaway. And the dwellers don't level up until they actually do something like succeed in a production rush or deal with a disaster. Anytime they do either they level up no matter how much XP they have.
